What is IgG?
The intact antibody of human immunoglobulin (IgG) includes the fragment for antigen binding (Fab) and the fragment that is crystallizable (Fc) for binding of Fcγ receptors.
IgG Subtypes
IgG has four subclasses or subtypes: human IgG IgG1, IgG2, IgG3, and IgG4, each with different constant regions and Fcγ receptor-binding affinities. When developing therapeutic antibodies, many factors are considered for IgG subtype selection.
Negative control
Regardless of which subclass or subtype of IgG is selected and the targeting antibody is developed, a high-quality negative control is necessary to characterize the antibody’s binding affinity of interest.
A clean and low-binding background from the negative control is critical. Nonspecific effects initiated by the antibody Fc backbone can complicate interpreting in vitro and preclinical results for antibody drug candidates (fully-human, humanized, or chimeric).
