The Background of cfDNA Extraction
In recent years, liquid biopsy has become a powerful alternative to tissue biopsy. Although biopsy is the gold standard to diagnose diseases (cell biology and molecular biology), biopsy is not always easy or possible to get. In these cases, liquid biopsy is useful for disease diagnosis, progression monitoring, and therapy responses. In these applications, cfDNA (cell-free DNA) and a small fraction of cfDNA-ctDNA (circulating tumor DNA) are often used as biomarkers in terms of cfDNA quantification, ctDNA mutation or methylation detection (qPCR, digital droplet PCR, or NGS).
For these applications, cfDNA is often required to be extracted from the blood samples before downstream analysis. However, the cfDNA level, especially in healthy individuals, is often very low. Extraction of cfDNA often leads to a big fraction of cfDNA loss, jeopardizing the accuracy and sensitivity of the clinical testing result. High-quality and high-yield cfDNA extraction become critical.
Manual cfDNA Extraction Workflow
Here is the workflow for manual cfDNA extraction. After plasma isolation, the two-step extraction by magnetic beads and column is applied to ensure the cfDNA extracted is pure and free of inhibitors on downstream PCR reactions.

Automated cfDNA Extraction Workflow
Unlike the manual cfDNA extraction kits, magnetic beads are also used for the second step of cfDNA extraction. The first- and second-step extractions are conducted on the automated extraction instrument MGISP-NE384.
